What it is

Kisspeptin is the hormone that sits at the top of the reproductive axis. Neurons in the hypothalamus release it, it switches on the GnRH neurons, and GnRH tells the pituitary to release LH and FSH, which drive testosterone, estrogen, sperm and ovulation. People born with broken kisspeptin receptors do not go through puberty. Kisspeptin-10 is the last 10 amino acids of the full 54-amino-acid hormone and is the smallest piece that still works.

In healthy men, a single intravenous dose raises LH about threefold within 30 minutes, and day-long infusions raised testosterone from about 16.6 to 24.0 nmol/L. In women the response depends on the cycle: it was weak or absent in the early follicular phase in one study and strong around ovulation and after menopause. A 2026 study kept LH up for 12 days in healthy men with daily 8-hour under-the-skin infusions, but a continuous 5-day infusion lost its LH effect. All of this comes from studies of 4 to 15 people run by a handful of academic groups.

Kisspeptin-10 is not approved anywhere. Online it is sold as a research chemical and promoted as a way to raise testosterone without shutting down the testes, or to protect fertility on testosterone therapy. Those uses have not been tested in trials. FDA placed kisspeptin-10 on its Category 2 compounding list in 2023 over immunogenicity and impurity concerns, and WADA prohibits it in male athletes.

Mechanism

How Kisspeptin-10 works

What it binds to, and what happens inside the body after that, step by step.

Outside the cell Inside the cell
Target Kisspeptin receptor KISS1R (GPR54), agonist

Kisspeptin-10 binds the kisspeptin receptor (KISS1R, formerly GPR54) on GnRH neurons in the hypothalamus. That releases GnRH, which drives the pituitary to release LH and, to a lesser degree, FSH. LH then tells the testes to make testosterone or the ovaries to make estrogen and support ovulation. Because it works upstream of the pituitary, the body's own feedback from sex hormones still shapes the response.

  1. 1 Receptor binding Kisspeptin-10 binds KISS1R on GnRH neurons in the hypothalamus.
  2. 2 GnRH release GnRH neurons fire, and in men infusions raised LH pulse frequency from 0.7 to 1.0 pulses per hour.
  3. 3 Pituitary response The pituitary releases LH, rising about threefold within 30 minutes of an IV dose in men, and smaller amounts of FSH.
  4. 4 Gonad response LH drives Leydig cells to make testosterone in men and supports estrogen production and ovulation in women.
  5. 5 Feedback and desensitization Sex steroids and contraceptives blunt the response, and continuous exposure for days can switch the LH response off.
The research

What the studies show

Human results and animal results are kept apart below. A result in rats tells you a mechanism is plausible. It does not tell you what happens in a person.

C Evidence

Small single-blind or open human physiology studies show kisspeptin-10 reliably raises LH and testosterone in men for hours, and in women depending on cycle phase. There are no randomized trials testing a clinical outcome such as fertility, symptoms of low testosterone, or libido with kisspeptin-10. The IVF and brain-imaging studies people quote online used kisspeptin-54, a different and longer-lasting form.

These are questions the published research has not answered.

  1. 01 Whether kisspeptin-10 improves any clinical outcome, such as sperm count, pregnancy rates, symptoms of low testosterone or libido, in a randomized trial.
  2. 02 Safety of daily dosing beyond 12 days.
  3. 03 Whether it preserves fertility during testosterone therapy; this has never been tested.
  4. 04 Bioavailability of a single subcutaneous shot, given the roughly 4-minute half-life.
  5. 05 Antibody formation with repeated injections.

Notes
Measured plasma half-life was 3.8 to 4.1 minutes in men and women, about 7 times shorter than kisspeptin-54. Because it clears so fast, most research used IV infusions or subcutaneous pumps rather than single shots. Continuous exposure for days can desensitize the LH response.

Groups with specific risks
  • Pregnancy: no human safety data
  • Hormone-sensitive cancers such as prostate or breast cancer: raises sex hormone levels, never studied in these groups
  • Competitive male athletes: prohibited by WADA
Interactions
  • Combined oral contraceptives: blocked the LH response to kisspeptin-10 in a small study of women.
  • Testosterone therapy and other sex steroids: suppress the reproductive axis by feedback and would be expected to blunt the response; not directly tested with kisspeptin-10.
  • GnRH agonists or antagonists (e.g., leuprolide, degarelix): act downstream on the same pathway; combination not studied.

Claims checked

What people say online, checked against the studies

Tap a claim to see what the studies found.

The claim"Kisspeptin-10 raises testosterone naturally without shutting down your own production the way TRT does."MixedSome studies agree, some do not

It does raise LH and testosterone for hours by working upstream of the pituitary. But continuous dosing for 5 days switched off the LH response, and no study has shown lasting testosterone gains in men with low testosterone.

The claim"Kisspeptin-10 keeps sperm production going while you are on testosterone."UntestedNobody has measured this in people

No study has given kisspeptin-10 alongside testosterone therapy and measured sperm. The evidence for preserving sperm on TRT comes from hCG, not kisspeptin.

The claim"Kisspeptin boosts libido and sexual attraction."UntestedNobody has measured this in people

A 2017 brain-imaging study in 29 men found more limbic activity in response to sexual images, but it used kisspeptin-54 and measured brain scans, not sexual behavior. No libido outcome has been studied with kisspeptin-10.

The claim"Kisspeptin is used to trigger ovulation in IVF."MixedSome studies agree, some do not

A 2014 study in 53 women used kisspeptin-54, not kisspeptin-10, to mature eggs, with a 23% clinical pregnancy rate. It is still experimental and not an approved IVF drug.

The claim"Kisspeptin-10 works the same in women as in men."Not supportedThe evidence does not back this

In one study women in the early follicular phase did not respond at all, while women near ovulation and after menopause did. Hormonal birth control blocked the response.

Untested pairings

Combinations people discuss

These pairings come up often. Almost none have been tested together in a trial, and the evidence note on each says so plainly.

Kisspeptin-10 with testosterone therapy

Promoted online to keep the testes active during TRT, in the way hCG is used.

EvidenceUntested. Testosterone suppresses the axis by feedback, and no study has measured sperm or testicular size with this combination.

Frequently asked questions
What is the difference between kisspeptin-10 and kisspeptin-54?
Both come from the same gene. Kisspeptin-54 is the main circulating form and lasts about 7 times longer in blood. Kisspeptin-10 is the shortest active fragment. Most of the IVF and brain-imaging research used kisspeptin-54.
Does kisspeptin-10 increase testosterone?
In healthy men and in men with type 2 diabetes and low testosterone, infusions raised testosterone over hours. In one study it rose from 16.6 to 24.0 nmol/L over 22.5 hours. Long-term effects in men with low testosterone have not been studied.
Is kisspeptin-10 FDA approved?
No. It is not approved anywhere, and since 2023 FDA has listed it under Category 2, meaning compounding pharmacies can face enforcement for making it.
How long does kisspeptin-10 last in the body?
About 4 minutes in plasma. LH rises within about 30 minutes of an IV dose and falls back over a few hours.
Is kisspeptin banned in sports?
Yes for men. WADA's 2026 list names kisspeptin and its agonist analogues as prohibited testosterone-stimulating peptides in males.
Can kisspeptin-10 treat infertility?
It has restarted hormone activity in a handful of women with low-GnRH conditions in exploratory studies, but no trial has shown it improves pregnancy rates.
