Peptides for longevity: what has been shown, and what has not

Nothing on this page has been shown to slow aging or add years to a human life. A few peptides change real outcomes in people with specific diseases. Most of the anti-aging claims come from cell dishes and mice. Here each one is mapped to the part of aging it is supposed to act on, with how far its proof actually reaches.

The short version
Nothing here has slowed human aging. No peptide has been shown to extend lifespan or slow aging in people. No trial has even tried to measure it.
One peptide changes hard outcomes. Semaglutide cut heart attacks, strokes and cardiovascular deaths by 20% in 17,604 people with heart disease and excess weight. That is a disease result, not proof of slower aging.
The anti-aging headlines come from mice. Epitalon, MOTS-c and FOXO4-DRI made old mice or cells look younger on some measures. Epitalon did not change average lifespan in mice, and the MOTS-c lifespan gain was not significant.
Growth hormone peptides push the wrong way. In animals, lower insulin and IGF-1 signaling goes with longer life. GH-releasing peptides raise IGF-1, and MK-677 raised blood sugar in older adults.
3 of 12 hallmarks have no peptide at all. loss of proteostasis, disabled macroautophagy, dysbiosis: no peptide in our database has been studied against them.
Most are research chemicals. 8 of the 16 compounds here have no approval anywhere and no active approval path.

Pulled live from the peptide database and grouped by evidence grade. Grades A to C have some human data; grade D has none. None of the human data comes from a trial of aging itself.

The biology of aging

Which part of aging each peptide is supposed to act on

Researchers group the causes of aging into 12 hallmarks, from DNA damage to failing mitochondria (López-Otín, Cell, 2023). The wheel colors each hallmark by the strongest study tying a peptide to it. Amber means the peptide pushes the pathway in the direction that shortens life in animal studies.

Mitochondrial dysfunction

Mitochondria make less energy and leak more damaging oxygen radicals.

  • NAD+Randomized human trials

Oral precursors raise blood NAD+ in trials, but a 2025 meta-analysis found no gain in muscle mass, grip or walking speed in older adults.

Growth hormone peptides push against the lifespan data

Less insulin and IGF-1 signaling

In mice, worms and flies, turning down insulin and IGF-1 signaling is one of the most repeated ways to lengthen life in the lab. In an Ecuadorian group of people who cannot respond to growth hormone, followed for 22 years, diabetes and cancer were almost absent compared with their relatives.

More growth hormone and IGF-1

This is what the GH peptides do. MK-677 raised IGF-1 to young-adult levels in older adults, and fasting glucose rose while insulin sensitivity fell. A hip-fracture trial was stopped early because of heart failure cases. Each profile lists the cancer risk of years of raised IGF-1 as unknown.

None of this proves that GH peptides shorten life. It means the case that raising growth hormone slows aging runs against the animal lifespan data, and no human trial has tested it either way.

What the evidence does not show

The claims you will see most often, next to what the cited study actually found.

The claim

"Epitalon extends lifespan."

What was found

In 108 mice, average lifespan did not change. Only the longest-lived 10% lived longer. There is no human lifespan data.

See the full profile
The claim

"Epitalon lengthens your telomeres."

What was found

It lengthened telomeres in cultured cells, including breast cancer cell lines in a 2025 study. Nobody has measured telomeres in a living person taking it.

See the full profile
The claim

"FOXO4-DRI reverses aging."

What was found

Old mice grew back fur and had better kidney function. In other mouse studies, clearing senescent cells raised lung blood pressure. It has never been given to a person.

See the full profile
The claim

"MOTS-c is exercise in a vial."

What was found

Old mice ran about twice as long. Their lifespan gain was not statistically significant. No human dosing trial has been published in a journal.

See the full profile
The claim

"An NAD+ drip recharges your cells."

What was found

In 8 men given an NAD+ infusion over 6 hours, blood NAD+ did not rise for the first 2 hours because it was cleared as fast as it went in. No controlled outcome trial exists.

See the full profile
The claim

"GH peptides restore youth."

What was found

MK-677 raised IGF-1 to young-adult levels and added lean mass in older adults, with no gain in strength or function, higher fasting glucose, and a trial stopped early for heart failure.

See the full profile
The claim

"Thymus peptides rejuvenate the immune system."

What was found

The largest thymosin alpha-1 trial, 1,106 people with sepsis, found no fewer deaths. No trial has tested it in healthy older adults.

One email when a longevity trial reads out or a regulator acts on a peptide. We read the study, not the press release, and say whether it was done in mice or people.

Frequently asked questions
Is there a peptide that is proven to extend human lifespan?
No. No peptide has been shown to extend lifespan or slow aging in people, and no trial has tested one for that purpose. The lifespan results people quote come from mice, and even there they are weak: Epitalon did not change average lifespan in a 108-mouse study, and late-life MOTS-c gave a 6.4% longer median lifespan that was not statistically significant.
Does Epitalon lengthen telomeres in humans?
That has not been shown. Epitalon switched on telomerase and lengthened telomeres in cultured human cells, including in an independent 2025 study that also saw lengthening in breast cancer cell lines. No controlled study has measured telomere length in the blood or tissue of people taking it. The human studies that exist are small, from the group that developed it, and do not describe randomization.
Is semaglutide an anti-aging drug?
It is a GLP-1 drug with strong outcome data in specific diseases: 20% fewer heart attacks, strokes and cardiovascular deaths in people with heart disease and excess weight (SELECT), and fewer kidney events and deaths in people with diabetes and kidney disease (FLOW). It has not been tested as an aging treatment in healthy, lean adults, and two large 2025 trials found it did not slow early Alzheimer’s disease.
Does IV NAD+ therapy work?
There is no controlled trial showing it improves energy, thinking, recovery or any aging marker. In an 8-person study, infused NAD+ was cleared from the blood as fast as it was given for the first 2 hours. Oral precursors (NR and NMN) do raise blood NAD+, but trials of them found no gain in muscle or function in older adults, and in a 410-person Parkinson’s trial NR made symptoms worse than placebo.
Are growth hormone peptides good for longevity?
The animal data point the other way. In mice, worms and flies, lower insulin and IGF-1 signaling goes with longer life. GH-releasing peptides such as MK-677, CJC-1295, sermorelin and tesamorelin raise IGF-1. In older adults MK-677 added lean mass without improving strength, raised fasting glucose, and a hip-fracture trial was stopped early because of heart failure. Whether years of higher IGF-1 change cancer risk is unknown.
What is FOXO4-DRI, and has it been tested in people?
FOXO4-DRI is a designed peptide that frees the protein p53 to kill senescent cells. In a 2017 mouse study it improved fur density and kidney function in old mice. It has never been given to a person in a published study, so there is no human dose or safety data. A 2023 study found that clearing senescent cells, including with FOXO4-DRI, worsened lung blood pressure in rodent models.
Is SS-31 (elamipretide) an anti-aging peptide?
SS-31 targets cardiolipin inside mitochondria and improved energy output and exercise tolerance in aged mice. In people it was FDA-approved in 2025 for Barth syndrome, a rare genetic disease, based on strength gains in about 10 patients without a placebo group. Its randomized trials in mitochondrial myopathy, heart attack and macular degeneration missed their main goals. It has not been tested in healthy aging.
