What it is

Selank is a synthetic heptapeptide made by taking tuftsin, a four-amino-acid fragment of an antibody protein that activates immune cells (Thr-Lys-Pro-Arg), and adding the same Pro-Gly-Pro tail used in Semax so it survives longer in blood. It came out of the Institute of Molecular Genetics in Moscow and was registered in Russia in 2009 as 0.15% nasal drops for anxiety, neurasthenia and stress-related disorders in adults. Since 2025 the Russian label lists it as nonprescription.

The human evidence is two Russian clinical studies. In 62 patients with generalized anxiety disorder or neurasthenia, Selank reduced anxiety about as much as the benzodiazepine medazepam, without the sedation. In 70 patients with anxiety disorders, adding Selank to phenazepam got the benzodiazepine working sooner and cut its side effects. Neither study was blinded against placebo in the way an FDA trial would be, and neither has been repeated outside Russia.

In the lab, Selank slows the enzymes that break down enkephalins (the body's own opioid peptides), shifts expression of GABA-related genes in cells and changes immune signaling molecules in blood. Which of these explains the calming effect in people, if any, is not settled. FDA placed Selank in its compounding Category 2 list in 2023 over immunogenicity concerns and lack of human safety data; the nomination was later withdrawn.

Mechanism

How Selank works

What it binds to, and what happens inside the body after that, step by step.

Outside the cell Inside the cell
Target Enkephalin-degrading peptidases (inhibitor, IC50 about 15-20 microM in human serum); GABA-A receptor binding (allosteric, rat brain membranes) and GABA-related gene expression; cytokine balance

Selank's calming effect is not explained by a single receptor. In human serum it inhibits the enzymes that degrade enkephalins, so these natural opioid peptides last longer. In cell and rat studies it changes expression of genes in the GABA system and of inflammatory cytokines. Radioligand work from the developers' institute in rat brain membranes suggests it changes GABA binding as a positive allosteric modulator at a site different from diazepam's. None of these mechanisms has been confirmed in people.

  1. 1 Nasal absorption Russian label data show 92.8% absolute bioavailability after intranasal dosing, with drug detectable in plasma within 30 seconds.
  2. 2 Enkephalins last longer Selank inhibits serum enzymes that break down leu-enkephalin; patients with generalized anxiety had a shortened enkephalin half-life at baseline.
  3. 3 GABA system genes shift In rat frontal cortex, Selank changed expression of 45 of 84 neurotransmission genes within 1 hour, including GABA receptor genes; a separate radioligand study found allosteric effects on GABA binding.
  4. 4 Immune signals change In patients treated for 14 days, the Th1/Th2 cytokine balance in serum shifted; in vitro it suppressed IL-6 gene expression in cells from depressed patients.
  5. 5 Fast clearance Plasma levels fall within about 5 minutes as tissue peptidases break it down; no intact drug appears in urine.
The research

What the studies show

Human results and animal results are kept apart below. A result in rats tells you a mechanism is plausible. It does not tell you what happens in a person.

C Evidence

Two Russian clinical studies (62 and 70 patients) found Selank reduced anxiety comparably to a benzodiazepine or improved benzodiazepine treatment when added on, but neither was a placebo-controlled, double-blind trial published in full in English. The mechanism work is mostly in rats and cells. There are no data in healthy people for focus, mood or sleep, which is how it is usually sold.

Gaps in the research

What nobody has measured yet

These are questions the published research has not answered.

  1. 01 Whether Selank beats placebo in a double-blind randomized anxiety trial.
  2. 02 Effects in healthy people on focus, mood, memory or sleep, which is how it is marketed online.
  3. 03 Human safety of injected Selank and of use beyond 14-day courses.
  4. 04 Which mechanism (enkephalins, GABA genes, cytokines) actually drives any anxiety effect.
  5. 05 Dependence or withdrawal with long-term use: never studied, though the short trials reported none.

Pharmacokinetics

How long it stays in the body

Half-life
Not established
Minutes in plasma (label: levels fall over about 5 minutes)
Routes studied
Intranasal (approved form), Subcutaneous injection (unapproved, gray-market use)
Bioavailability
92.8% absolute bioavailability after intranasal dosing, per the Russian label
Notes
The Russian label reports plasma drug within 30 seconds of nasal dosing and a fall in concentration over 5 to 5.5 minutes, with no intact drug or metabolites in 24-hour urine. These are manufacturer data; no independent human PK study has been published in English. Nothing is published on injected Selank in humans.

No human half-life has been published for Selank, so we do not draw a clearance curve. Half-lives quoted online for it are usually estimates from animal data.

Claims checked

What people say online, checked against the studies

Tap a claim to see what the studies found.

The claim"Selank works like a benzodiazepine without the downsides."MixedSome studies agree, some do not

One Russian study found anxiety relief similar to medazepam without sedation, but it was small and not placebo-controlled. Long-term dependence has not been studied.

The claim"Selank boosts focus and memory in healthy people."UntestedNobody has measured this in people

No published trial measured cognition in healthy adults. The rat memory studies and a brain-scan study do not show this.

The claim"Selank is a GABA agonist."Not supportedThe evidence does not back this

It is not shown to activate GABA receptors directly. Rat studies from the developers' institute show changes in GABA-related gene expression and an allosteric effect on GABA binding in brain membranes. None of this has been measured in humans.

The claim"Selank strengthens the immune system."MixedSome studies agree, some do not

It is derived from an immune peptide and shifted cytokine levels in anxious patients, but no study shows fewer infections.

The claim"Selank is FDA-approved for anxiety."Not supportedThe evidence does not back this

It is registered only in Russia. FDA has never approved it and it cannot be legally compounded in the US.

Frequently asked questions
Is Selank legal in the US?
It is not FDA-approved and is not on the list of substances pharmacies may use for compounding. It is sold online as a research chemical, so purity and dose are unverified.
Does Selank help anxiety?
Two Russian studies of 62 and 70 patients suggest it reduces anxiety about as well as a benzodiazepine or improves benzodiazepine treatment. Neither was a placebo-controlled double-blind trial, so the effect size is uncertain.
Is Selank addictive?
The short Russian studies reported no withdrawal, but dependence over months of use has never been studied.
Semax vs Selank: which is for what?
Selank is built from tuftsin and is registered for anxiety. Semax is built from an ACTH fragment and is registered for stroke and cognitive problems. Neither has Western trial data.
How fast does Selank work?
The label reports drug in plasma within 30 seconds and gone within minutes. The clinical studies measured anxiety over days to two weeks, not minutes.
Can you take Selank with antidepressants or benzodiazepines?
It was studied added to phenazepam, a benzodiazepine, in one Russian trial. No study has looked at SSRIs or other psychiatric drugs, so ask a prescriber.
