What it is

Growth hormone is a 191-amino-acid protein. In the early 1990s researchers at Monash University found that a short piece from its far end, residues 177 to 191, kept growth hormone's fat-burning effect in rodent fat tissue without its effects on growth or blood sugar. Adding a tyrosine to the front of that piece produced AOD-9604. Products sold as HGH Fragment 176-191 usually carry the same 16-residue sequence as AOD-9604, so for practical purposes the two names describe one compound.

In obese mice and rats the fragment cut weight gain, raised fat burning and did not cause the high blood sugar that full growth hormone does. It does not bind the growth hormone receptor and does not raise IGF-1. Those animal results led to six placebo-controlled human trials in about 900 people, run by the developer, Metabolic Pharmaceuticals.

The human trials showed the drug was well tolerated, with side effects similar to placebo, no rise in IGF-1 and no harm to glucose tolerance. They did not show it works. The largest, a 24-week trial in 502 obese adults, did not produce significant weight loss compared with placebo at 12 or 24 weeks, and the developer ended the obesity program in February 2007. Those trials used oral tablets and IV infusions. The daily under-the-skin injections people use today have never been tested in a published trial.

Mechanism

How HGH Fragment 176-191 works

What it binds to, and what happens inside the body after that, step by step.

Outside the cell Inside the cell
Target Unknown receptor on fat cells; does not act through the growth hormone receptor

Animal and cell work suggests the C-terminal piece of growth hormone acts on fat cells to raise fat breakdown and lower fat storage. It does not bind or activate the growth hormone receptor, does not raise IGF-1, and in mice did not raise blood sugar. Its actual receptor is unknown; it raised beta-3 adrenergic receptor expression in obese mice. In mice lacking that receptor, two weeks of dosing no longer cut weight, but a single dose still raised fat burning.

  1. 1 Reaches fat tissue After injection or infusion the fragment circulates to adipose tissue; how much survives after oral dosing is unclear.
  2. 2 Raises fat breakdown In rodent and human fat tissue samples it increased lipolysis, the release of stored fat as glycerol and fatty acids.
  3. 3 Cuts fat storage It lowered lipogenesis, the building of new fat, in rodent fat tissue.
  4. 4 Skips growth signals It does not bind the growth hormone receptor, does not raise IGF-1 and did not cause cell growth or high blood sugar in mice.
  5. 5 Weight effect in animals only Obese mice and rats gained less weight. Human trials did not reproduce a significant weight loss.
The research

What the studies show

Human results and animal results are kept apart below. A result in rats tells you a mechanism is plausible. It does not tell you what happens in a person.

B Evidence

Grade B reflects that randomized human trials exist, not that they were positive. Six company-run placebo-controlled trials of the AOD-9604 sequence in about 900 people showed good tolerability and no effect on IGF-1 or blood sugar, but the decisive 24-week trial found no significant weight loss and was never published in a journal. Those trials used oral and IV dosing; the injected product sold today has no human data. The fat-burning effects are shown in rodents and in fat tissue samples only.

Gaps in the research

What nobody has measured yet

These are questions the published research has not answered.

  1. 01 Whether subcutaneous injection, the way it is used today, causes any fat loss in humans.
  2. 02 Half-life and absorption after injection.
  3. 03 Long-term safety beyond 24 weeks.
  4. 04 The receptor it acts through.
  5. 05 Whether reported weight results from the 12-week trial hold up, since efficacy data were never published in a peer-reviewed journal.
  6. 06 Identity and purity of products sold as HGH Fragment 176-191.

Pharmacokinetics

How long it stays in the body

Half-life
Not established
Unknown in humans. The human trials did not report a half-life.
Routes studied
Oral tablet or capsule (human trials), Intravenous infusion (human trials), Subcutaneous injection (animal studies and unregulated use)
Bioavailability
Not reported for any route in humans.
Notes
In human serum and urine samples in the lab the peptide breaks into several fragments; one, CRSVEGSCG, is much more stable than the parent and is used to extend detection in doping tests. No human pharmacokinetic study of subcutaneous injection has been published.

No human half-life has been published for HGH Fragment 176-191, so we do not draw a clearance curve. Half-lives quoted online for it are usually estimates from animal data.

Claims checked

What people say online, checked against the studies

Tap a claim to see what the studies found.

The claim"HGH Fragment 176-191 burns fat without the side effects of growth hormone."MixedSome studies agree, some do not

The side-effect half is supported: trials found no rise in IGF-1 and no harm to blood sugar. The fat-burning half is not: the largest human trial found no significant weight loss over 24 weeks.

The claim"It is the part of growth hormone responsible for fat loss, so it works like growth hormone for fat."Not supportedThe evidence does not back this

That was the hypothesis from rodent fat tissue. When it was tested in obese adults, the fragment did not produce significant weight loss.

The claim"HGH Fragment 176-191 and AOD-9604 are different compounds."MixedSome studies agree, some do not

The native 176-191 sequence starts with phenylalanine, but most products sold under this name carry the tyrosine-modified AOD-9604 sequence. Unless a supplier shows otherwise, treat them as the same molecule.

The claim"It targets belly fat specifically."UntestedNobody has measured this in people

No human study measured fat distribution. The claim is borrowed from growth hormone research.

The claim"It helps repair cartilage."UntestedNobody has measured this in people

One rabbit osteoarthritis study used joint injections. No human data exist.

Untested pairings

Combinations people discuss

These pairings come up often. Almost none have been tested together in a trial, and the evidence note on each says so plainly.

HGH Fragment with CJC-1295 and Ipamorelin

Marketed as pairing a fat-targeting fragment with peptides that raise the body's own growth hormone.

EvidenceNo study has tested this combination. The fragment alone failed its human weight-loss trial.

Frequently asked questions
Is HGH Fragment 176-191 the same as AOD-9604?
Usually, yes. AOD-9604 is growth hormone residues 177 to 191 with a tyrosine added at the front, and that is the sequence most products labeled 176-191 contain. The true native 176-191 piece starts with phenylalanine instead of tyrosine.
Does HGH Fragment 176-191 actually work for fat loss?
Not in the human trials that exist. The largest, 24 weeks in 502 obese adults, did not produce significant weight loss compared with placebo, and the developer stopped the program. Fat loss has been shown in obese mice and rats.
Does it raise IGF-1 or affect blood sugar?
No. In human trials it did not change IGF-1 levels or glucose tolerance. That was its main selling point over full growth hormone.
Is HGH Fragment 176-191 legal?
It is not an approved drug in any country. In the US it cannot be used in pharmacy compounding: an FDA advisory committee voted 12 to 0 in 2024 against adding AOD-9604 to the compounding list, and FDA lists it with safety concerns. WADA bans both AOD-9604 and hGH 176-191 for athletes.
Has injecting it been studied?
Not in people. The human trials gave it by mouth or by IV infusion. Subcutaneous injection has only been used in animal research.
